Abstract :

Adenoviruses are potentially useful vectors for vaccination and gene therapy. An extensive purification is done during the production of recombinant adenoviruses in order to obtain the pure final products. However, product-related impurities, such as virus aggregates and virus incomplete particles, can be present in the final product and may lead to adverse effects or lack of efficacy. The goal of this study was to obtain a suitable analytical method for determining the purity of adenovirus based on the content of product-related impurities. RP-HPLC was used to differentiate between the complete and incomplete particles based on the protein profiles. The mobile phase was (A) 0.1 % TFA in 5% acetonitrile and (B) 0.1% TFA in 99% acetonitrile. The column and autosampler temperatures were maintained at 45oC and 4oC, respectively. The flow rate was 0.2 mL/min with gradient program. The chromatograms were monitored at 280 nm. The proposed method can be used for determining the purity of adenovirus.
