Abstract :

The objectives of this research were to obtain phenylpropanoic acid derivatives and to approve their activity as antiplatelet. Phenylpropanoic acid and the methoxy substituted of phenylpropanoic acids (ortho and para positions) were  synthesized  through  catalytic hydrogenation of  the  vynilic double bond of phenylpropenoic acid derivatives.  The reaction  was carried  out  at room temperature using Raney-Ni  in  NaOH condition.  The structure of synthesis products were identified by FTIR and 1H-NMR spectroscopy. The yield of phenylpropanoic acid, 3-(4-methoxyphenyl)propanoic acid, 3-(2-methoxyphenyl) propanoic acid were 74%, 61%, 44% respectively.  Their activity as antiplatelet was confirmed  through in silico study  into active site of cyclooxygenase-1 (pdb. 1CQE).  The all compounds showed no significantly differences ligand-enzyme interaction energy than acetylsalicylic acid. So, our research concluded that compounds can be used as antiplatelet like acetylsalicylic acid through inhibition of cyclooxygenase-1.
