Effect of protease inhibitors on in-vitro growth of fusobacterium nucleatum atcc 25586

      Abstract :

      Fusobacterium nucleatum has been implicated in the pathogenesis of periodontal diseases. It has been reported that F. nucleatum can obtain energy from the fermentation of certain amino acids, free or in the form of peptides. From nutritional aspects, the question arise is wether the growth of F. nucleatum in both carbon and energy sources will be affected by protease inhibitors. The aim of the present study was to investigate the effect of protease inhibitors on the growth of F. nucleatum ATCC 25586 in both brain heart infusion (BHI), consisting mainly of peptides and a chemically defined medium (CDM), containing free amino acids as the sole source of carbon and energy. The organisms were grown anaerobically in either BHI and CDM, then protease inhibitors tested were added to both cultures and incubated at 37o C. Optical density readings were taken about 3 h intervals up to 24 h. Protease inhibitors under investigation were able to depress the organism’s growth in the BHI. Incontrast, they showed little or no inhibitory activity towards CDM-growth.
