The difference between polyclonal antibody titer from 185 kda and 67 kda streptococcus mutans

      Abstract :

      Streptococcus mutans has some antigenic structures in its cell wall, one of them is protein antigen which has the different molecule weight such as 67 kDa and 185 kDa. From the result of various studies, it shows that the protein antigen has nature of antigen as the caries pathogenesis. The objective of this study was to get polyclonal protein from the cell wall of S. mutans, especially the protein with molecule weight 67 kDa and 185 kDa, which could be used as an immunization agent in preventing tooth caries. And to answer the problem if there is difference of antibody titer in the animals tested which were immunized polyclonal antigen 67 kDa and 185 kDa. The study was done in stages and it was started with S. mutans isolation from the plaque of tooth caries patient. The S. mutans colony in which its cell wall was extracted, and then it was tested with electrophoresis to get the immunogenic protein with molecule weight 67 kDa and 185 kDa. The protein result which was eluted was used as the immunization agent in the animal test. The immunization was done 4 times with 14 day interval, then from the blood serum it was titrated with ELISA to know the IgG value in each rat in each group. The result of ELISA titration in this study showed that the IgG value of control study which was not immunized had relatively consistent value, and there was an increasing of IgG value was higher in all rats of the group which was immunized polyclonal protein 185 kDa than the group immunized 67 kDa.
