Abstract :
In an attempt to separate X and Y bearing chromosome spermatozoa using several techniques, a study has been designed to evaluate the effect of the treatment itself to the number of blastomeres of the embryos resulted 48 hours after an in vitro fertilization of these sperm to eggs. Frozen thawed Madura bull semen were treated in six different methods to separate X and Y bearing chromosome spermatozoa by using sephadex, percoll, Swim up , Swim up + Aside migration, direct current electricity 1,5Volt and control group. Ovaries were collected from an abattoir and subsequently oocytes were matured in a TCM199 media with a supplement of 10% estrous cow serum in a 5% C02 incubator for 24 hours. Fourty eight hours after the in vitro fertilization using the sperm collected from these separation methods, the percentage of embryos consisted of more than 4 blastomeres and that of less than 4 blastomeres were determined. The result showed that the effectiveness technique on separating the spermatozoa Y bearing chromosome were Swim up , combination of Swim up and Aside migration, and anode electricity 1,5 Volt (p
