Abstract :

The aim of this research was to identify Staphylococcal enterotoxin of Staphylococcus aureus isolated from Swiftlet’ nest. Indonesia has a lot of natural resources, one of them is Switflet’snest, as a food. It consist of 17 essensial amino acids, minerals, and any vitamins. Nowdays, it is founded Staphylococcus aureus in Switflet’snest. But not all of Staphylococcus aureus cause staphylococcal switflet’snest poisoning. Only Staphylococcus aureus strain enterotoxigenic produce enterotoxin. The characteristics of enterotoxin are resistant of 100ºC heat for 30 minutes, proteolytic enzyme resistant and superantigen, which can stimulate lymphocyte subpopulation directly and fast (1-6 hours) which will cause gastroenteritis, emetic, nausea, and vomit.The result of this experiment showed that Staphylococcus aureus strain enterotoxigenic D and P on number 9B. Detection of Forward Sequences was based on DNA Sequencing analysis result was related to the sep gene. The result of phenotype detection depicted that DNA fragment is able to ligated to pMOSBlue-T vector. Measurement of the length plasmid recombinant increases ± 3500 basepairs. After transformation to Escherichia coli DH5α, plasmid recombinant has T7 Promoter and being supported by RNA polymerase of Escherichia coli DH5α was capable to express enterotoxin, its ± 26 kDa weight molecules. To confirm an expression of enterotoxin D was done by latex agglutination is based on with antigen-antibody reaction which was specific D.
