The differences concentration of tooth whitening material toward cytotoxicity using mtt assay

Abstract :
ABSTRACT The aim of this study was to evaluate the cytotoxicity of several tooth whitening material, carbamide peroxide 10%, 15%, 20% and hydrogen peroxide 38% toward BHK-21 cell using MTT assay. Each well of microplates which used for the test were aliquotted BHK-21 cell suspension, after that the test solution were added to eight well each group, respectively. Microplates were incubated at 37°C in 5% CO2 incubator for 20 hour. MTT stock was added to each well, incubate again for 4 hours, after that DMSO was added. The microplate were detected using ELISA reader at 630 nm. The cytotoxicity was evaluated by the 50% cytotoxic dose (CD50), which was determined by calculating the proliferation percentages of cells treated with various concentrations of tooth whitening gel, and comparing the results with that of the positive and the negative control. The result showed that percentages of the living cell at 10% carbamide peroxide group = 86,73%; 15% = 81,22%; 20% = 81,82%; 38% hidrogen peroksida = 64,08%, respectively. Anova test and LSD showed no significant difference between 10%, 15%, 20% groups, but there are significant toward 38% group and controle. The 38% hydrogen peroxide group is expectable to be more cytotoxic than those containing 10% carbamide peroxide. which is equivalent to 3.6% hidrogen peroxide. Hydrogen peroxide releases oxygen. Exposure to higher concentrations of hydrogen peroxide can increased the oxidative reactions and damages by free radicals. The oxidative reactions and subsequent damage in cells are the major mechanisms responsible for the toxicity of peroxide compounds. Conclusion. The10%, 15%, 20% carbamide peroxide and 38% hydrogen peroxide tooth whitening agents conclusion were not detected cytotoxic toward BHK-21 cell line using MTT assay within CD50.
