The effect of catechins green tea toward p-110 phosphatidil inositol 3- kinase ( pi3k ) expression and p38 mitogen activated protein kinase (mapk ) activity in aortic male rats induced high fat diet

Abstract :

Catechins, isolate from green tea  GMB4 clones could be developed as a potential preventive agent for atherosclerosis. This activity is thought to be based on the structure epimerisation, hydroxy phenolic  and galloil group of these isolates. Therefore the purpose of this study is to prove whether catechins ,isolate from green tea,  inhibits the decrease of p110-PI3K expression and  the increase of p38-MAPK activity in male wistar rats induced by  high  fat diet. The method used in this study were Western Blott and ELISA to determine  p110-PI3K expression and  activity of p38 MAPK, respectively. The results showed that  catechins inhibited significantly (p<0.05) the decrease of the expression of p110 PI3K at dose of 6 mg/day and 24 mg/day,but not in the dose 3mg/day. This suggested that dose 6mg/day and 24 mg/day was able to repair the damage signaling insulin on PI3K pathway caused by insulin resistance. While the dose  3 mg/day of catechins  inhibited the increase activity of p38 MAPK. These could be  based on the ability of catechins as antioxidant. Catechins with hydroxy phenolic group could be  a potential electron donor to bind  the free radicals. Polyphenol catechins may also modulate  signaling cellular processes, such as NFkβ, activator protein-1 DNA binding, glutathione biosynthesis, PI3K/Akt, MAPK pathway and the Nrf2 transcription factor that regulates expression of several antioxidant genes. It can be concluded that catechins  inhibits  the decrease of p110 PI3K  (at  dose of 6 mg/day and 24 mg/day) and also the increase of  p38 MAPK activity (in all treatment doses).
