Abstract :

Diagnosis to Leucocytozoonosis by blood and organs smear examinations is to identify gamet and schizont stages, respectively. However, those diagnostic techniques are mostly late coincided severe infection and death of host. Based on the reason, serological diagnostic is really needed to establish the accurate diagnostic. The aims of the research are to identify immunogenic protein of Leucocytozoon schizont then to be continued to produce specific polyclonal antibody. The specific polyclonal antibody could be used as moleculer diagnostic kitt. Moreover, immunogenic protein of Leucocytozoon schizont could be used as vaccine. Schizont protein of pulmo, liver kidney and spleen were isolated by sonication technique. The protein fraction was identified by SDS-Page with electrophoresis. Then, the proteins were administrated to rabbit to produce polyclonal antibody and to those polyclonal antibody were assigned immunogenic protein by immunoblotting. The result of the research showed that the Leucocytozoon has six protein fractions to molecular weight. They were 155.8, 48.98, 39.81, 37.15, 35.48, 30.20 and 23.99 kDa respectively. Among those protein fractions, 48.9, 39.8, 37.2 and 35.5 kDa were immunogenic protein that could be produced on rabbit.
