The using of brucella abortus s-19 lipopolysaccharide as diagnostic antigen for detection of antibody by elisa technique

      Abstract :

      The aim of this study was isolated and identified the character of Brucella abortus LPS. By identification of Brucella abortus LPS character that base on immunogenecity and antigenecity, therefore Brucella abortus LPS could be established as diagnostic antigen. The experiment methods were (1) cultivation Brucella abortus S19 on Trypticase Soy Agar (TSA) and Potato Agar (PA), (2) Isolation and LPS extraction by phenol, (3) polyclonal antibody production by immunizing rabbit and mice with whole bacterial cell of Brucella abortus S19, (4) LPS characterization by indirect sandwich-ELISA technique. The results showed that (1) Brucella abortus LPS could be separated from cell- wall by phenol extraction (2) LPS characterization obviously had highly antigenecity but low immunogenecity. Therefore Brucella abortus LPS could be used as diagnostic antigen to the antibody examination by indirect ELISA technique.
