Effect of different duration administration of testosterone on insulin resistance index and free fatty acid concentration. an experimental study on pcos in rats.

Abstract :

The objective of this study was to confirm the increase in insulin resistance index and free fatty acid concentration after administration in different duration of testosterone propionate and placebo in Rattus Novergicus strain wistar as an PCOS model. This study was performed at animal test facility and Pathology Clinic Department in Veterinary Faculty, Airlangga University, November 2007 until January 2008 period, as approved by Animal Research ethics committee, Veterinary Faculty, Airlangga University. This was an experimental research with randomized post test only control group design. Sample are consecutively recruited based on inclusive criteria and divided into two groups. The interventional group were stimulated with testosterone propionate for 14, 21 and 28 days. Concentration of glucose were measured by glucose oxydase method, meanwhile insulin and free fatty acid were measured by ELISA method. Insulin resistance index was measured with HOMA. There were significant increases in insulin resistance index between interventional and control groups. Statistically insulin resistance index on serum rat interventional group is significantly increase from control group in 14 days administration (3.7933 ± 0.5416 : 0.1800 ± 3.5350, p< 0.05), in 21 days administration (7.4950 ±  2.5289 : 13.2017 ± 4.6473, p< 0.05), and in 28 days administration (8.2950 ± 2.5403 : 20.1100 ± 9.8162, p< 0.05). Statistically free fatty acid concentration on serum rat interventional group is significantly increase from control group in 21 days administration (0.4467 ± 0.1199 : 1.1033 ± 1.6224, p< 0.05), and in 28 days administration (0.545 ± 0.2471 : 2.265 ± 1.7539, p< 0,05) but not in group 14 days administration. In conclusion, insulin resistance index on serum rat interventional groups is significant increase from control group. Free fatty acid concentration on serum rat interventional group is significant increase from control group in 21 and 28 days administration.
