Abstract :

Infection with enterotoxigenic Escherichia coli (ETEC) is one of the main causes of neonatal calf diarrhea in developing countries. The major virulence factors of enterotoxigenic strains of Escherichia coli in neonatal is also the heat-stable enterotoxin (ST) and K99 pili proteins. In a case control study Esherichia coli isolated from faecal samples from dairy calves were phenotyped by biochemical fingerprinting and analyzed for virulence genes by PCR. PCR assays, which are more rapid, sensitive, and specific, have also been widely used for ETEC diagnosis. This research used non probability sampling. The samples taken from 10 dairy farmers placed in Malang who have cases of death in calves and poor sanitation criteria. Samples were firstly plated on Eosyn Methylene Blue Agar (EMBA), and afterwards the colonies that suspect Escherichia coli (colored metallic green) confirmed by Indol test, the other biochemistry test, and Blood Agar (BA) enrichment media. Positively test, later was  tested by PCR by using primer ST and primer K99. Results of this research showed that all 10 samples of suspect Escherichia coli on media EMBA and 7 of the 10 positive samples in Indol test and other Biochemistry test. Results of electrophoresis of PCR products from 7 positive samples of Escherichia coli, 3 samples showed positive for the presence of genes encoding ST amplicon 147 bp in length. While the results of PCR using primers K99 nothing positive. Based on the test result  ETEC in the samples may not indicate the presence gene encoding K99 pili proteins.
