Abstract :

Semen is the liquid or semigelatinous cellular suspension containing the male gametes orspermatozoa and secretions from accessory organs of the male reproductive tract. The fluid portion of this suspension, which is formed at ejaculation, is known as seminal plasma. Seminal plasma is composite secretion arising from a number of sources including the testes, epididymis, and accessory glands of the male. Seminal plasma appears to be more important in natural mating of cow where the ejaculate is deposited in the vagina. Seminal plasma contains of citric acid, ergothionine, fructose, glycerylphosphorylcholine and sorbitol. Appreciable quantities of ascorbic acid, amino acids, peptides,proteins, lipids, fatty acids and numerous enzyme. Seminal plasma was reported to be important for maintaining spermatozoa motility, improving sperm viability and increasing the resistance of boar spermatozoa to cold shock damage.Protein of plasma seminalis can be isolated by protein purification with added Phosphat Buffer Saline (PBS) and Phenylmethenesulfonyl fluoride (PMSF) and then centrifugated. The supernatan added by etanol absolut and finally to measure protein total concentration by using Biuret method. The result of this experiment show that the protein total concentration was 8200 ppm/cc plasma seminalis.
