Abstract :

The general objective of the research are to characterize genotypes of Viral Nervous Necrosis (VNN) and analyze kinship genotypes on grouper in East Java and West Nusa Tenggara. The samples of this research were taken from the fish center of the cultivation of various regions, such as Situbondo region, Banyuwangi, Gresik, Tuban, West Lombok, East Lombok and Sumbawa. This research is divided into two stages, the first step is by inspecting of grouper which are indicated VNN infection with the tecnique of conventional PCR and the second step is following by amplification using specific primer coat protein and DNA sequensing. After that followed by homology analysis and then do a comparison with the data of China RGNNV EF558369 GenBank and Bali HQ859926.1. From the result of research shows that the point mutation at nucleotide base pair which has been compared to the data from China RGNNV EF558369 GenBank and Bali HQ859926.1. The Kinship analysis is based on the nucleotide composition between regions which has the same kinship that is included in the genotype Red-spotted Grouper Nervous Necrosis Virus (RGNNV) in which after the comparison between regions have similarities ranging from 9593.53%-98.63% with the data GenBank GenBank Gondol BALI HQ859926 and EF558369.1 RGNNV China. Phylogenetic analysis based on the grouping of each sample seen that each area has in common with the genotype of the reference data RGNNV Isolate china and different from the other genotypes. This analysis serves to determine kinship because of the distribution of seeds from one area to another.
