Abstract :

The contain protein in mammal, especially, human sperm membrane is , 50%, that distributed astrans membrane protein. The protein membrane of human sperm have any function as transport system,protein carrier and receptor or ligand and have numerous antigenic determinants including carbohydrate and protein, namely glycoprotein, that can be corfirmed by PAS detection. This study focused on the identification of proteins membrane from human sperm based on its ratio of protein, carbohydrate and total glycoprotein. Confirmation of protein concentration measured Biuret method and the ratio glycoprotein;carbohydrate and protein measured by Periodic Acid Schiff (PAS) reagent, which detected aldehyde groupin sugar produced by action of periodic acid. The molecular weight of glycoprotein membrane of humanspermatozoa was detected by showing through the use of SDS-PAGE. The result showed that protein band with molecular weight of 97, 32 and 27 kDa that contained glycoprotein 25,52; 28,91; 26,17 mg/ml,carbohydrate of 10,84; 11,97 and 10,57 mg/ml and protein of 14.96; 16,94 and 15.34 mg/ml, respectively.It can be concluded that the ratio glycoprotein; carbohydrate and protein of human sperm membrane are 2:1:1; 2:1 :1 and 2.5:1:1,6, for glycoprotein with molecular weight of 97, 32 and 27 kDa, respectively.
