Abstract :

This research aims to improve the quality of post-thawing of frozen semen, with the addition of specific protein osteopontin spermatozoa membrane into a freezing medium. This research was conducted in the laboratory and in the field that includes the stages following methods frozen semen quality test supplemented osteopontin spermatozoa membranes isolated cow. The test includes: measuring the levels of osteopontin using the standard osteopontin ELISA, determination of the optimum dosage of the addition of osteopontin in freezing media on the quality of spermatozoa post thawing, which include : the percentage of motility, viability, plasma membrane intact and acrosome intact hood. Results showed that the addition of specific protein osteopontin membrane of the spermatozoa in the semen freezing diluter media there are significant differences (p < 0.05) compared with no addition of osteopontin this means that the addition of specific protein osteopontin membrane can increase the number of spermatozoa motility, viability, plasma membrane intact (MPU). The conclusion from this study is the addition of a specific protein osteopontin spermatozoa membrane can improve the quality of frozen semen and improvement of in vitro fertilization.
