Abstract :
Thin layer chromatography-densitometry for determination of colchicine in Gloriosa superba Linn. infuse has been validated using selectivity, accuracy, precision and linearity parameter. The aim of this study was to obtain a valid method for determination of colchicin in Gloriosa superba Linn. infuse. The results were as follow, selective mobile phase was chloroform: diethylamine (9:1). Rf value of colchicine was 0.35 and it was completely separated from another compound in sample with Rs of 2.14 and 3.5, respectively. Linearity test showed there was linear relationship between colchicine concentration and area, represented by coefficient of correlation (r) of 0.9965 and Vxo of 8.19%. The average colchicine recovery was (85.77±6.39) % and its precision was 7.45%. RSD for instrument precision was 3.74%. The results of this study indicated that this method was valid and met the requirement of AOAC. Applying this method, the colchicine concentration obtained in Gloriosa superba Linn. infuse was 0.73 ppm.
