Abstract :
In this work, tempeh of Canavalia ensiformis that fermented by Rhizopus oryzae FNCC 6078 was used as a source of fibrinolytic enzyme. This enzyme could provide clinical application for blood clot removal. That fibrinolytic enzyme was extracted by blending and liofilization. The crude enzyme extract was assaying in fibrin plate agar showing fibrinolytic activity. Partial purification of the crude enzyme extract was performed by two steps, i.e., by ammonium sulphate fractionation followed by gel filtration chromatography using sephadex G-100 as stationary phase. The fibrinolytic activity were evaluated at each step of purification. Fibrinolytic enzyme obtained from 50-70% ammonium sulfate fraction had the highest spesific activity 6,6 x 10-2 U/mg; 10,0 x 10-2 U/mg; dan 23,8 x 10-2 U/mg. There was no significant difference between spesific activity of sample before and after partial purification.
