Abstract :

Formalin is a commercial name of formaldehyde solution 35-40% in water. Formalin kills virus, bacteria, fungi and parasite and has found wide use as a disinfectant with a broad efficiency. Formalin is often used as a food preservative with concentration range from 1,88–413,89 ppm (mg/kg), although it is not permitted since formalin is toxic for human consumption. Therefore the requirement of a chemical reagent for the examination of formalin in food is very necessary. The aim of this research is validating Spectrophoto-metric method to detect formalin in chicken meat.This research is conducted to develop method to detect formalin in chicken meat by reaction with chromotropic acid in strongly acidic media producing violet-red colour followed by detection with Spectrophotometric method at 567,0 nm. The research shows that the method is able to detect formalin in chicken with limit of detection is 0,0058 ppm and limit of quantitation is 0,0192 ppm. The standard curve gives regression y = 0,5233x+0,0397, r value is 0,9998 (p<0,01). Precision study shows  Coefficient Variation 0,4039%. Recovery study shows (87,3934±1,0826)%. The samples of chicken collected from a traditional market in Surabaya are detected concentration formalin range from 74,7595-140,2042 ppm (µg/g; wet weight).   Keywords : validation,Spectrophotometric, formalin, chicken meat, chromotropic acid.
