The role of 5α reductase inhibitor (finasteride) in prostate

Abstract :

This was an experimental study using post-test control group design involving Wistar strain Rattus norvegicus as experimental animals. The objective of this study was to elaborate the action mechanism of finasteride in the prostate. Samples were divided randomly into 2 groups, i.e., 1 month- and 2 month-groups, each comprising 26 rats. Each group was further divided into 2 subgroups, i.e., control and treatment (finasteride) groups, each consisting of 13 rats. After treatment for 1 month and 2 months, the prostate from each rat was removed for TGF ß-1, EGF, and FGF examinations using immunohistochemistry and proliferative degree examination using graticulae. Univariate analysis was used with t-2 sample test since TGF ß-1, EGF, and FGF had no correlation. The analysis showed that finasteride could significantly reduce TGF ß-1, EGF, FGF and proliferation, both in 1 month and 2 monthgroups. Conclusively, this study had successfully elaborated the action mechanism of finasteride in the prostate. It was found that finasteride reduced TGF ß-1, EGF, FGF and proliferation, so that the volume of the prostate was reduced and BPH symptoms relieved. However, the reduction of the TGF ß-1, EGF, FGF and proliferation was not maximal, although it was statistically significant. The author suggests another study in which finasteride and anti-estrogen therapies are applied to prostatic tissue to maximally reduce TGF ß-1, EGF, FGF and proliferation, so that they may restore BPH symptoms.
