Abstract :

O. gouramy cultivation was in Indonesia, moreover central of cultivation not only in Java but non Java. One of cultivation constraint is production decrease caused by bacterial. A. hydrophilla cause of O. gouramy disease. This disease become an epidemic on 1-2 week in fresh water cultivation with high mortality about 80-100%. Vaccination is more effective measure to prevent bacterial disease. Vaccination can increase immune system on fish body to specific pathogen and suppress mortality. The research to determine different dose whole cell A. hydrophilla effect of survival rate, total leucocyte O. gouramy, and optimum dose of vaccine. Research method used an experimental test with Completely Randomized Design, six treatment with three replication. There are two control and four different dose, A (control -), B (control +), C (105 cell/ml), D (106 cell/ml), E (107 cell/ml), and F (108 cell/ml). Main parameter are survival rate of fish, total leucocyte of fish and clinical sign. Second parameter is water quality of cultivation. Survival rate of fish was calculation after finished. Total leucocyte was calculation in day-0, day-7, day-14, and day-21. Monitoring of clinical sign during 7 days after challenge test. Based on the result of research, different dose of vaccine have significant effect (p<0,05) to survival rate and total leucocyte of fish. The highest of survival rate of fish in treatment D 54,17%. The highest total leucocyte in treatment D (20,43x103 cell/mm3). Treatment D with dose vaccine 106 sel/ml was given optimum result on survival rate and total leucocyte of fish, although total leucocyte between treatment D, E, and F not significant but the highest survival rate on treatment D. Clinical sign of fish control more appear than fish which vaccination.
