Optimasi campuran manitol - pemanis stevia sebagai pemanis tablet kunyah ekstrak jahe merah (zingiber officinale roscoe var.rubrum) dengan metode simplex lattice design

Abstract :

Red ginger has activity as antimotion sickness, therefore it should be developed into chewable tablet dosage form. Formulation of chewable tablet need sweetener in order to increase acceptablity.The aim of the research is to know the optimum composition of manitol and stevia as sweetener in formulation of red ginger tablet with Simplex Lattice Design method. Red ginger extract was obtained by using maceration method with 70% ethanol as solvent. Chewable tablets made from red ginger ethanol extract by dry granulation method were manufactured in three formulas based on simplex lattice design; formula I (100% mannitol), formula II (50% mannitol and 50% stevia), and also formula III (100% stevia). Tablet was evaluated physical properties of tablet: hardness, friability, taste and uniformity of weight. Data was evaluated to get SLD equation to determine the best composition. SLD equation validation was performed by t-test with 95% level of confidence, between response of physical properties of tablet base on equation and experiment result. The results showed that the increasing proportion of stevia would increase the hardness and decrease the friability and the weight uniformity of the tablets. Optimum formula was obtained in 50% manitol and 50% stevia with value 12.66% for granule tapping index, 0.085 g/ml for granule absorption, 8.97kg for granule compactibility, 0.935%CV for tablet weight uniformity, 9.46 kg for tablet hardness, 0.523% for tablet friability and 50% of tablet taste responses said the tablet was acepptable. Based on this study, the combination of filler mannitol and stevia (50%: 50%) is able to produce the optimum formulation with good weight uniformity, hardness and friability and also acceptable taste responses by the respondents.
