The production of superoxide from shrimp penaeus monodon hemocyte as an stress indicator on the intensive culture

Abstract :
The experiments were deal to understand the production of superoxide from shrimp hemocyte P.monodon after and before stimulated with Vibrio bacterin zymosan and ECP. The shrimp hemocyte have a phagocytic activity against bacteria or other foreign substance, but the activity will disturbed if shrimp under stress. The production of superoxide was low if shrimp reared under stress condition compared to shrimp reared under normal condition. The OD value hemocyte of shrimp reared under heavy density induced by zymosan is 0.52, Vibrio sp 0.46 and ECP 0.45. Meanwhile OD value shrimp reared without stress condition are 1.2 induced by zymosan, Vibrio sp. 1.19 and ECP 1.18. The OD vale of control fish are 0.61 induce by zymosan, 0.6 Vibrio sp. bacterin and ECP the lowest of 0.55. PI value of phagocytic hemocyte was relatively low after 1 and 3 days stress treatment are 2.8 and 1.9 while the control are 1.5 and 1.3. The low value of PI due to the decreased of phagocytic function of hemocyte. Based on the results,the production of superoxide is influenced by the stress, therefore to keep the shrimp under low stress is very good aquaculture management.
