Abstract :
This research was conducted to investigate the effect of separation X and Y bearing chromosome spermatozoa using several techniques on the percentage of live and motility of sperm, and cleveage rate after in vitro fertilization using its sperm. Frozen thawed Madura bull semen were treated in six different methods to separate X and Y bearing chromosome spermatozoa by using sephadex, percoll, Swim up, Swim up + aside migration, electric current 1.5 Volt and control group. Ovaries were collected from an abattoir and subsequently oocytes were matured in a TCM199 media with a supplement of 10% estrous cow serum in a 5% C02 incubator for 24 hours. Forty eight hours after the in vitro fertilization using the sperm collected from these separation methods, the percentage of cleavage was determined. The result showed that the percentage of live sperm after treatment using Sephadex and Percoll column were not significantly different (p>0.05) compared to control group.
