Effect of glycerol in skim egg yolk diluent on the freezing of goldfish spermatozoa (cyprinus carpio)

Abstract :

Glycerol plays an important role in the processing of frozen semen. Glycerol is one of the intracellular cryoprotectants, which is able to replace the water in the cell and shift the electrolytes out of the cell. Egg yolk as extracellular protection provided by lipoproteins forward to protect spermatozoa from cold shock during the freezing process by stabilizing the sperm membrane. The aims of his study to define the best concentration of glycerol in the skim egg yolk diluents for the motility and viability of carp (Cyprinus carpio) spermatozoa pre-freezing and post-thawing. Glycerol concentrations were used 11%, 13% and 15% in diluter B skim egg yolk. This research using statistical test with completely randomized full designed by six replicants and had been analyzed by ANOVA (analysis of variance) and continued with Honestly Significant Difference test (HSD) at a significance level of 5%. The results showed that the use of different concentrations of glycerol had no effect on carp sperm motility and viability. The highest average percentage obtained with 15% glycerol concentration was 53.83 ± 17.497 for motility, and 67.50 ± 17.026 for viability. The lowest motility and viability were obtained with 11% glycerol was of 49.16 ± 9.174 for motility and 63.66 ± 7.339 for viability.
