Abstract :

The aim of this study was designed to determine the characteristic of dosage form, released and penetration of diclofenac sodium with w/o microemulsion system from HPMC 4000 gel. Microemulsion contained ratio surfactant Span 80-Tween 80 and cosurfactant isopropanol 4:1. The concentration of diclofenac sodium was  5% (Hendradi et al, 2012). There were two formulas in this study, a gel with w/o microemulsion system as the first formula and gel with w/o emulsion system as the second formula. The characteristic evaluation included appearance, pH, and ability to spread diameter of zero load. Formula I had a better stability than formula II and broken after 4 days formulation. Then, only formula I can be evaluated by penetration test. The data of characteristic studay was analized by statistic using independent t-test with degree of confident 95%. The released  and penetration study of diclofenac sodium was carried out with Erweka Dissolution Tester Type DT 820 with apparatus 5-paddle overdisk in phosphate buffer saline 7.40±0.05, temperature 37.0±0.5oC, 100 rpm. As membrane released study was cellophane, and Wistar rat skin was used as membrane at penetration study.The result was showed that preparation of microemulsion system was significant different on pH, consistency, and spreadibility compared with  the emulsion system with degree of Freedom 95% (α = 0,05), but it was same  effect on color and odor. The result showed that drug released rate was 51.79±0.60 µg/cm2 /min1/2, penetration rate for formula I was  0.71 ± 0.03 µg/cm2/min and the membrane Wistar rat skin permeability for formula I was 7.26x10-5 ± 3.49x10-6 cm/min.
