Abstract :
Dengue virus (DENV) is a single stranded RNA virus that circulates in the body of primate and causes DHF human. Dengue virus have four serotypes (DENV-1, DENV-2, DENV-3, DENV-4). Four dengue virus serotypes do have a similar, yet distinct antigenic properties. Infection only one serotype will give immunity to one serotype is concerned, but does not provide cross- immunity. This study aims to determine the immunogenicity of dengue virus (DENV-1, DENV-2, DENV-3 and DENV-4) inactive as a candidate dengue cocktail vaccine. This study used 18 mices were divided into three treatment groups, P0 immunized 0.5 cc PBS, P1 immunized with cocktail of dengue virus (DENV-1, DENV-2, DENV-3 and DENV-4) inactive as much as 0.5 cc TCID50107, P2 immunized cocktail of dengue virus (DENV-1, DENV-2, DENV-3 and DENV-4) inactive as much as 0.5 cc TCID50108. At day 0,7,14 and 21 days of 18 mice that had been treated blood drawn to determine the levels of immunoglobulin M and G with indirect elisa method. On day 28, the treatment group P1 and P2 in the challenge test with a cocktail immunized with dengue virus (DENV-1, DENV-2, DENV-3 and DENV-4) active 0.5 cc TCID50108 for the presence of viremia. The results of this study are significant differences (p<0.05) between the groups P0 (control), P1 and P2 in terms of inducing titers of immunoglobulin (IgM, IgG, IgG1a, IgG2a, and IgG2b). In the challenge test using active dengue virus (DENV-1, DENV-2, DENV-3 and DENV-4) TCID50108 in experimental animals (P1 and P2), then the PCR test followed by electrophoresis results showed the absence of viremia or viral dengue as evidenced by a negative PCR result.
