Comparison of sperm quality after separation x and y chromosome between column albumin method and electric separating sperm (ess) method on ekor gemuk sheep

Abstract :

This aims of this study was to determine quality of the sperm (motiltias, vitality, and integrity of membranes) spermatozoa and the size of the sperm head after undergoing a process of separation of sheep chromosome X and Y with two different methods, namely albumin column method using blood serum and Bovine Serum Albumin sheep (BSA) and electric separating sperm (ESS) methods. The study consisted of 6 treatments, namely P0, P1, P2, P3, P4, P5 and each treatment consisted of ten replicates. Data obtained on the quality and size of the sperm head of sheep were analyzed and statistically tested using ANOVA followed by Tukey 5% significance. The results showed that the blood serum of sheep and BSA did not significantly affect the quality of spermatozoa sheep, the most tangible result is shown by the control treatment. Size X and Y spermatozoa head on P1 and P2 showed significant differences to the P0, P1 ratio on chromosome X and Y spermatozoa was 28%: 72% and 32% in comparison P2: 68%. In the second method, namely electric separating sperm (ESS), the results showed that the quality of the sperm cells in test microscopic observation of motility in all treatment groups showed no significant differences, while the power of life and abnormalities observation results show that the results between the P3 and P4 control do not have significant differences but significantly different from P5
