Effect of seminal plasm of cow on goat’s spermatozoa plasma membran in milk diluter on semen freezhing

Abstract :
Up till now semen freezing process of ram’s spermatozoa has not yet satisfying, this is because ram’s seminal plasma contain phospholipase A from bulbourethral gland, that can coagulate with egg yolk in diluter. Because of that researchers want to replace ram’s seminal plasma with bull’s seminal plasma in milk diluter media. Special aim of this research is to understand the benefit of bull’s seminal plasma in diluter of ram’s spermatozoa. General purpose of this research is using bull’s seminal plasma in diluter of ram’s spermatozoa to improve fresh semen quality and even frozen semen. This research consists of three treatments. Control treatment (PO) are diluter + ram’s semen (without seminal plasma). Treatment I (PI) are diluter + spermatozoa + bull’s seminal plasma (1:1), treatment II (PII) are diluter + spermatozoa + bull’s seminal plasma (1:2). All of treatments observed everyday of motility, viability, plasma membran integrity and spermatozoa’s DNA integrity. Observation in diluter without seminal plasma shows lowest spermatozoa’s membrane plasma integrity in day I, II, III, IV, and V. Anova test for spermatozoa’s membrane plasma integrity shows significant different (p <0,05) between PO, PI, PII, and PIII. Duncan's multiple range test for observation PI group produce highest spermatozoa’s membrane plasma integrity percentage.In conclusion, addition of bull’s seminal plasma can mantain, membrane plasma integrity in skim milk diluter
