Evaluation toluidine blue staining to identify connective tissue mast cells (ctms) in paraffin block thin skin of dog

Abstract :
Mast cells have an important role in allergic diseases because it contains histamine, it is the mediator of allergic reactions and stored in granules. Identification of mast cells can be done with some staining, for example by Alcian blue staining, safranin, berberine and immunohistochemical techniques. Various staining techniques is often done, in terms of a long process requiring a relatively long time, therefore it is necessary to find an alternative staining method that is easy to do, does not require a long time and cheap. This study aims to determine the effectiveness of the use of Toluidine Blue staining for identification of mast cells in the thin skin tissue of dog. Dog thin skin tissue (two years old, Male Labrador Reriever) used were obtained from the collection of tissue stored in Histology Laboratory School of Veterinary and Biomedical Science Murdoch University with a case code no. H12-944 in 10% formalin fixation fluid. Tissue processed for paraffin blocks were made and then sliced into two series, each series was repeated to 10 slides. The first series of thin skin tissue stained with routine staining techniques (Hematoxylin-Eosin Harris) according to the protocol in Bancroft and Gamble (2008), while the second series stained with Toluidine Blue staining. Results of tissue that has been stained and then observed under a microscope with 400x magnification for identification of mast cells. Data were analyzed by independent t test (5% significance) to determine whether there are differences between the two series of staining (Hematoxylin-Eosin and Toluidine Blue staining).The result of this research indicate that the use of routine staining methods (Hematoxylin-Eosin) and Toluidine Blue staining to identify mast cells in the skin thin dogs significantly (P <0.05) showed differences, with an average ± standard deviation of the number of mast cells were identified respectively is 9.85 ± 1.31 and 18.64±2:19. Toluidine Blue staining method can be used as an alternative of special staining to identify mast cells, it is easy and does not require an expensive cost.
