Identification of cyclin-dependent kinase (cdk1) that express on bovine cumulus oocyte complex after a model oocyte maturation experimence

Abstract :

CDK1 is the major protein that influence in oocyte maturation. Oocyte maturation not only influence by the hormonal but also by growth factor. This study is a series of experiments on CDK1 protein fraction in the process on in vitro maturation for increasing quality of oocytes in the in vitro embryo production. Bovine oocyte used as a oocyte maturation model. The objective of this study to identify CDK1 from follicles with diameter 3-8 mm. The follicles of bovine ovaries obtained from slaughterhouse were aspired. The in vitro maturation oocytes were lysed by sonification. Identification of CDK1 protein fraction was performed with SDS-PAGE. The isolate protein injection to rabbits to get the antibody of CDK1. The spesifity of the antigen was performed with Dot Blot method. From the result of this experiment, several protein band have appeared. 7 protein fraction obtained : 163,25 kDa; 131,15 kDa; 94,43 kDa; 71,82 kDa; 56,14 kDa; 36,23 kDa dan 28,32 kDa. Protein band with 32 kDa molecular weight was identified as Cyclin Dependent Kinase (CDK1). From the spesifity test used the Dot Blot method obtained the good positive result were after second booster. However, further study with various follicles diameters are needed.
