Identification of kit-ligand from in vitro maturation of bovine oocytes with electrophoresis method

Abstract :
The expression patterns of Kit-Ligand and c-kit in the ovary, as well as the actions of Kit-Ligand on oocytes and theca cells, suggest that Kit-Ligand important for many stages of follicular development in the ovary. The aim of this experiment was to identify and characterize the Kit-Ligand protein fraction that isolated from mature oocytes of bovine ovaries obtained from all folikel. The folikel of bovine ovaries obtained from slaughterhouse were aspired. The oocytes were matured in Tissue Culture Medium 199 (TCM 199) then culture for 22 hours at 38,5 ºC in incubator CO2. Identification of Kit-Ligand protein fraction was performed with Sodium Dodecyl Sulphate Polyacrilamid Gel Electrophoresis (SDS-PAGE) process. From the result of this experiment, saveral protein band have appeared. Based of the calculation using regression equation on the protein marker for estimating the molecular weight of Kit-Ligand ,12 protein fractions were obtained : 193,09 kDa, 172,51 kDa, 105,84 kDa, 89,37 kDa, 64,94 kDa, 53,81 kDa, 39,84 kDa, 36,27 kDa, 30,63 kDa, 26,35 kDa, 22,68 kDa, 20,64 kDa. Protein appearing in the protein band the molecular weight of 36,27 kDa was identified as protein Kit-Ligand that has the important role in the follicle development.
