Influence of leaf extract of sirih ( piper betle l.) to growth staphylococcus aureus and escherichia coli with disk diffusion method

Abstract :
Bakterisidal effect had been known in sirih plant ( Piper betle L.). This research was to know bakterisidal effectand the effectivity of leaf extract of sirih ( Piper Betle L.) based on in-vitro test to bacteria of Staphylococcusaureus and Escherichia coli. This Research had used diffusion methode that sensitivity knowledgeable Kirby-Bauer test with five treatment and seven times restating. Five treatment were consisted of negatif control 10 %DMSO, leaf extract of sirih was 2,5; 5, 10% and antibiotic as positif control. Paper disk sterile soaked in 10%DMSO and in leaf extract of sirih with concentration of 2,5; 5 and 10% respectively. Paper disk have beensoaked to be evaporated in incubator till the disk to dry, then put down at media Mueller Hinton Agar that hasbeen inoculated Staphylococcus aureus with oxacillin and also Escherichia coli with sulfonamide, then incubationat temperature of 37°C for 24 hours. The result of research shown at concentration 2,5; 5 and 10% ofleaf extract of sirih and also antibiotic can inhibited growth of Staphylococcus aureus and Escherichia coli.With double distance test of Duncan indicate that highest result of antibacterial effect of leaf extract of sirih toStaphylococcus aureus is by concentration at 10% that not different significant with concentration of 2,5 and5%, while antibiotic of Oxacillin show lower result than third concentration of leaf extract of sirih. Highestresult Escherichia coli shown by sulphonamide antibiotic, comparated with third concentration of leaf extractof sirih. Control 10% DMSO don’t have effect to both bacteria.
