Protein characterization of insulin like growth factor – i complex of goat seminal plasma as an capacitation medium of sperm

Abstract :

Nineteen percentage of goat semen consist of seminal plasma. Seminal plasma reported to be important for maintaining sperm motility, sperm viability and for increasing the resistance to cold damage. Seminal plasma, a physiological secretion from multiple glands of the male reproductive tract, is the natural medium for maturation, enzymatic and surface-modifiing events. Seminal plasma contains a variety of biochemical components some of which are relatively specific for the regulation of spermatozoa function. The addition and removal of variety of protein during epididymal maturation and at ejaculation play important roles in the capacitation of sperm and fertilizationof the egg. In this article it will be discussed about the protein characterization of goat seminal plasma by Sodium Dodesil Sulfat – Poliakrilamid Gel (SDS-PAGE) 12% procedure. Fractination of goat seminal plasma protein by SDS-PAGE procedur would be result seven fractions. First fraction is with molecul mass 150,288 kDa, second fraction is with molecul mass 103,486 kDa, 3rd fraction is with molecul mass 76,155 kDa, 4th fraction is with molecul mass 53,249 kDa, 5th fraction is with molecul mass 35,378 kDa, 6th fraction is with molecul mass 14,099 kDa and 7th fraction is with molecul mass 11,492 kDa. The further demonstrate the first fraction with molecul mass 150 kDa is ternary protein complex consist of a molecul IGFBP-3 (53 kDa), a molecul IGF-I or IGF-II (7,5-10 kDa) and unique acid labil subuinit/ALS (76 kDa). In conclution of this study is goat seminal plasma protein component consist of seven fractions and that it’s could be to maintain the sperm.
