Spesifisity test with dot blotting of epidermal growth factor (egf) isolated from cumulus oocyte complex after in vitro maturation

Abstract :
The aim of this study was to know the character of epidermal growth factor (EGF) protein isolated from bovine oocyte. Bovine ovary was collected from slaughter house washed with NaCl. An aspiration technique was used to collect oocyte. Those oocytes were maturated by in vitro condition then the EGF protein was isolated after running in SDS PAGE. Polyclonal antibody was produced by sub cutaneous injected of crude protein oocytes containing EGF in rabbit, then followed by dot blotting examination. Booster was done twice after 3rd and 7th bleeding then continued until 10 times every week. The antibody anti EGF from rabbits sera was reacted with EGF antigen. The dot blot showed, the intensity of dot color increased significantly from 4th bleeding and reaches the highest concentration of antibody at 9th bleeding. It meant, bovine oocyte EGF protein could be characterized by dot blotting, of which the highest antibody titer detected at eleventh week after sub cutaneous injection of crude protein oocytes containing EGF.
