The effect of alkaloid fraction of jarong (achyrantes aspera linn) on viability culture myeloma cell mice

Abstract :

The purpose of this research is to prove that the effect of alkaloid fraction of jarong (Achyrantes aspera linn)can be used as a cure for cancer with invitro on cultured cell of myeloma cell of mice using the method of cellcounting. In this research, the percentage of the perceived cell viability of the living myeloma cell is comparedwith the dead cell using the negative and positive control. This research divided into five group: P0 is controlnegative,P1 liquid test 1 ppm, P2 liquid test 10 ppm, P3 liqiud test 100 ppm, P4 liquid test 1000 ppm and P5positive control of 100 ppm colchisin. All groups were repeated four times and incubated for 24 hours in anincubator 95% O2 5% CO2 at temperature of 37 OC. Then the culture which had been treated was taken 0,1ccand give 0,1cc of 4% tripan blue. Later the dead cells and the living cell were calculated using the thomamethod with the help of hemositometer. The living cells will be colorless and dead cell will be tinted.The resultof assasment of the myeloma cells using hemositometer showed that the cultured cell in the concentration of100 ppm and 1000 ppm will have lower viability compared with the concentration of 1 ppm, 10 ppm and positivecontrol. In the concentration 1 ppm, the alkaloid fraction of jarong (Achyrantes aspera linn) killed more than 50percentage of myeloma. The alkaloid fraction of jarong (Achyrantes aspera linn) on the viability of myeloma cellculture of mice will have Lethal Concentration 50 : 0,716 ppm.
