The effect of various egg duck concentration in skim milk as semen diluters of fat-tailed sheep for motility, viability and intact plasma membrane spermatozoa before freezing

Abstract :

This study aimed to prove the effect of duck egg yolk in skim milk against motility, viability, integrity of plasma membrane of Domba Ekor Gemuk sperm before freezing. This study use Domba Ekor Gemuk semen with additional skim milk diluter which divided into four groups and each group contain six times repeat. Those groups are control group (K) which is semen with skim milk added with chicken egg yolk 5%, treatment group (P1) is semen with skim milk added with duck egg yolk 2,5%, treatment group (P2) is semen with skim milk added with duck egg yolk 5%, treatment group (P3) is semen with skim milk added with duck egg yolk 7,5%. The variables which observed are motility, viability, and the integrity of plasma membrane. Collected data were tested with Kruskal Wallis test dan if there is a different continued with Mann Whitney test. The results of this study show that there is significant differences between chicken egg yolk diluter with semen using duck egg yolk diluter (p<0,05). The best semen using skim milk diluter with addition of duck egg yolk 5% which is spermatozoa motility 74,1%, sperm viability 84,4% and sperm plasma membrane integrity 80,4%, so that those semen can be used as frozen semen.
