The effects of insulin transferin selenium (its) supplementation on embryo freezing media at morula stage towards the percentage of embryonic development at blastula stage

Abstract :
The aim of this research was to evaluate the influence of Insulin Transferin Selenium (ITS) Supplementation on embryo freezing media at morula stage towards the percentage of embryonic development at blastula stage. The experimental animals used female mice strain Balb C which were development at blastula stage. The experimental animals used female mice strain Balb C which were superovulated by using Pregnant Mare Serum Gonadotropin (PMSG) and human Chorionic Gonadotropin (hCG), afterwards monomatting was carried out. Three days after vaginal plug examination, the mice were sacrificed by conducting dislocatio cervicalis, next the uterus was removed and flushed. The morula embryos were classified into three groups : without ITS, ITS with 5μg/ml and 10μg/ml of concentration. Each treatment group was put into ministraw 0,5 ml and stored into liquid nitrogen for a week then thawed immediately. The post-thawed embryos were cultured by using tissue culture medium for 24 hours. Based on the statistical test, it demonstrated that there were no significant differences between the treatment group p>0,05with Chi square value of 1,144. Nevertheless, if it was investigated based on the percentage calculation of the blastula embryo development, it proved that 10μg/ml of ITS could increase the percentage of morula embryo development into blastula by 54% compared to 37% without ITS. In conclusion, based on statistical analysis, the distribution of Insulin Transferin Selenium (ITS) supplementation concentration on freezing media had not functioning optimally in increasing the morula embryos development into blastula after being cultured.
