Difference in the productivity of b-lymphocyte isolated from lymph nodes and that isolated from mice spleen in the production of toxocara cati monoclonal antibody using hybridoma technology

Abstract :

Monoclonal antibody is highly required, particularly in diagnostic development. The antibody can be produced through hybridoma technology. Hibridoma is a result of fusion between myeloma and B-lymphocytes that has been sensitized by an immunogene. Today, the B-lymphocyte used for producing hybridoma is the one isolated from spleen. However, the obtained formation of hybridoma is remarkably less. One method to increase the productivity of monoclonal antibody production is by using B-lymphocyte isolated from lymph nodes. The difference in productivity between B-lymphocyte isolated from lymph nodes and that isolated from spleen remains unclear. This was an experimental study using 32 culture cells, in which myeloma cells were fused with B-lymphocyte from lymph nodes and that from spleen of mice that had been sensitized with embryonated egg of T. cati intraperitoneally. Obtained results revealed significant difference (p < 0.05), either in the number of formed hybridoma or its capability to produce antibody, between hybridoma resulting from fusion of myeloma and B-lymphocyte isolated from lymph nodes and that from isolated from spleen. In conclusion, B-lymphocyte isolated from lymph nodes is more productive than that isolated from mice spleen as the pair of myeloma cells in the production of monoclonal antibody using hybridoma technology.
