Abstract :
Dental pulp therapy of teeth degraded by caries, dental wear lesions or trauma in currently limited to conventional restoration such as pulp capping or root canal therapy. Therefore, dental research focusing on strategies for vital pulp therapy by maintaining the function of the teeth. The implementation of a medical device in dental and oral tissue engineering is based on potensial cell sources and biocompatible materials applied as direct pulp capping agents, wich can also serve as a carrier of signaling molecules in addition to nerves and blood vessels, the pulp contains highly prolifrative stem/progenitor cells possessing a self renewal and differentiation capability. Methods. These researches were in-vitro in fibroblast tissue culture of BHK and in-vivo from orthodontic patients indicated for premolar extraction, between ages 10 – 15 years. A class V cavity preparation was created in the buccal aspect 1 mm above gingival margin until pulp exposure. Cavity was irrigated slowly with saline solution and dried with a sterile small cotton pellet. The sterile absorbable collagen membrane used, as inert carrier of TGF-1 was soaked with 5 l. TGF-1 was covered by a teflon pledge to separate pulp capping agent from restoration of Glass Ionomer Cement. Results. In-vitro there was 30ng/ml of TGF-1 was highest absorbance in spectrophotometer, and in-vivo was occur inflammation in pulp fibroblast. In further clinical trial experiment there was 20ng/ml not occurring inflammation symptom. Conclusion. In-vitro experiment of exogenous TGF-1 on BHK tissue culture only for estimating to clinical trial, and the same consentration of TGF-1 applied to human fibroblast cause inflammation symptoms.
