Uji spesifisitas protein prolaktin dari serum darah itik molting melalui metode western blot

Abstract :

The aim of this study was to isolated and identified prolactine hormone from molting duck’s serum using western blot method. The result of this study could use as a scientific base to produce the antiprolactine polyclonal antibody that could accelerate the molting process. The serum taken from blood at the axillaris vein was added SAS 50%, homogenated, sentrifugated, and dialyzed. Prolactin characterization started by SDS PAGE to determine the molecule weight. Specificity test by western blot and electroelution to get the pure prolactin isolated. The result of this study showed that prolactin protein could be isolated from the molting duck’s serum by SAS 50% method. The identification of the prolactin protein by SDS PAGE showed the prolactin at 27 kDa molecule weight. The specificity test by western blot and electroelution can be used to obtain that the isolate was pure prolactin isolate with 27 kDa molecule weight
