Antigenecity, sensitivity and spesificity of 27-28 kda protein of fasciola spp excretory-secretory (es) material for distomatosis diagnosis in cattle sera by using indirect-elisa technique

Abstract :

This research was aimed to find out antigenic protein with high sensitivity and specivicity for producing a diagnostic kit of distomatosis through blood test of patients. Isolation and identification of protein originated from ES of Fasciola spp were focused to get P27-28 kDa. In the first step mature of Fasciola spp adult were incubated in RPMI medium for producing ES protein. The second step, the protein fractions were determinated to carry out based on MR result and then this protein was isolated by preparative gel electrophoresis. The third step, the specific protein was identified by SDS-PAGE with silver stain. The fourth step, tests of sensitivity and specificity for CatL protein which was isolated in the second step and these procedure were also carried out for ES protein which was isolated in the first step. The result showed: 1) It was isolated various proteins, that were proteins in the molecular weight (MW) 40, 35, and 27-28 kDa and 2) As a material for diagnostic test, P27-28 kDa with the points 63.6% of sensitivity and 87.5% spesificity was better than ES protein with the points 100% of sensitivity but its sensitivity was 0%.
