Abstract :
The aim of this research was to analyze adaptation capability and antigen stability of Newcastle Disease (ND) virus at HeLa cell line. ND virus was inoculated at primary cell CEF and HeLa cell line with two different dose (100μl and 200μl). The virus growth was observed every day to 72 hours and ND virus was inoculated (blind passage) until CPE reached 80% at both kind of culture cell. Two kind of sample, ND virus from both infected culture cell medium and virus from infected cell was collected and prepared for the next step. The measuring of the titer was done by HA micro technique test. Sample with highest titer of HA micro technique test then analyzed by SDS-PAGE and Western Blot method. The result showed that ND virus inoculated at primary culture cell CEF and HeLa were both adapted well, signed by cytophatic effect such as monolayer growth disturbance and the forming of synctia. Meanwhile as the result of ND virus protein identification by SDS PAGE, 4 band was obtained from virus that infected HeLa cell and from ND virus of the culture fluid obtained 12 band. ND virus adapted at HeLa cell by Western blot method showed that ND virus has reactivity against chicken anti ND serum. This proved by 2 protein band formed by virus extraction to HeLa cell and 6 protein band by culture fluid HeLa cell.
