Abstract :

The aim of this research was to find out the reactivity of Brucella abortus S-19 outer membrane protein toward rabbit polyclonal antibody which have been injected with Brucella abortus S-19. In advanced it will be used as diagnostic kit that has sensitivity and highly specific for Brucellosis diagnosis.Brucella abortus S-19 was obtained from PUSVETMA bacteria stock. The suspention consist of Brucella abortus S-19 and the adjuvant was injected subcutaneous into rabbit in order to get polyclonal antibody anti Brucella abortus S-19. Booster has been done for six times.OMP was obtained from Brucella abortus S-19 by sonication technique, using ultrasonic homogenizer. The identification of immunogenic antigen protein fraction had done by western blot. Western blot technique used OMP antigen and polyclonal antibody (anti Brucella abortus S-19).The result of this research showed that OMP Brucella abortus S-19 have had five fractions. These protein had recognized antibody with molecul weight of : 55,3 kDa, 24,4 kDa, 21,9 kDa, 20,3 kDa and 14,7 kDa.
