The effectiveness of giving haemopoetic stem cells in the pregnant mice (mus musculus) exposed to plumbum towards the decreasing number of necrocis cells and expression of caspase-3 in placenta

Abstract :
The aim of this study to observe the effectiveness of Haemopoitic Stem Cell in pregnant mice exposed to plumbum, and the observed cases are the numbers of necrosis cells and caspase-3 expression in the placenta. Haemopoetic Stem Cells was expected to able to reduce the number of necrosis cells and was abled to reduce the expression of caspase- 3. This study used a randomized design, and the required samples in this study were 40 female mice and 40 male mice (Mus musculus). Each treatment utilized 10 male mice and 10 female mice with details as follow, (1) first treatment group (T1): pregnant mice were given a dose of plumbum 1/12 LD50 (54.42 mg / ml) during gestation days 9-15. (2) second treatment group (T2): pregnant mice were given a dose of plumbum 1/16 LD50 (40.75 mg / ml) during gestation days 9-15. (3) Third treatment group (T3): Plumbum pregnant mice were given a dose of 1/12 LD50 (54.42 mg / ml) during gestation days 9-15 and on day 9 Hemopoetic Stem Cell 1x107 was given intravenously. (4) fourth treatment group (T4): pregnant mice were given a dose of Plumbum 1/16 LD50 (40.75 mg / ml) during gestation days 9-15 and on day 9 Hemopoetic Stem Cell 1x107 was given intravenously. The results of the expression of caspase-3 between treatments are significant, especially the fourth treatment group (T4). Of all the treatments (T1, T2, T3) when compared to treatment P4 shows a significantly different result. T4 was treated by giving Hemopoetic Stem Cells on day 9 which coincided with the first day of plumbum exposure. Intended stem cells function properly because the homing process is good enough for stem cells.
