The potential of soursop (annona muricata linn.) leaves extract as immunostimulantin rabbit pbmc

Abstract :
The usage of plant-based tradisional medicine is getting popular because there is minimum risk to health unlike chemical or synthetic drugs. One of the alternative usage of herbal medicine as immunostimulant to increase immune system is by using soursop leaves (Annona muricata Linn.). Soursop leaves extract contains flavanoid from flavonol group which can improve activation and proliferation of T-cell. The aim of this research is to prove that soursop leaves ethanol extract with dosage 5 μg, 15 μg, 25 μg and 50 μg have immunostimulant potensial to increase the expression of T-cell and secretion IFNγ in PBMC White New Zealand rabbits. The result from immunocytochemistry for expression on White New Zealand rabbit PBMC culture shows the lowest dosage 5 μg of soursop leaves extract is able to stimulate the expression of T-cell (47.64±0.7021) which greatly differs from control group (K), comparative control Phylantus niruri extract (P0), and experimental group P2, P3 and P4 (p ≤ 0.05). The increased number of T-cell indicates that flavonol in soursop leaves helps to activation macrofag to release IL-12 and TNFα, cytokine TNFα induces NK cell to release IFNγ, IFNγ induces T-cell . The test for IFNγ amount secretion with ELISA method on White New Zealand rabbit PBMC culture show that at the lowest dosage 5 μg soursop leaves ethanol extract is able to increase the amount of secreted IFNγ (353.89±54.04) which greatly differs from control group (K), and experiment P2, P3 and P4 (p ≤ 0.05), but not significant difference from comparative control with Phylantus niruri extract (P0). The increase IFNγ secretion proves that glycoside flavonol in soursop leaves ethanol extract induces thymocyte to increase IFNγ transcription.
