Abstract :
Penelitian ini bertujuan mengembangkan suatu metode deteksi asam nukleat yang non-radioaktif, sederhana, sensitif dan cepat terhadap strain Escherichia coli yang menghasilkan toksin VT (Verocytotoxin), yaitu dengan mengoptimasikan kondisi reaksi probe oligonukleat dan meningkatkan efisiensi berbagai langkah yang terlibat dalam preparasi DNA, hibridisasi langsung pada gel agarose kering, labeling dan kualitas DNA yang dihasilkan.
Hibridisasi probe yang non-radioaktif pada gel agarose kering cukup sederhana dan memberi hasil yang baik, tidak diperlukan blotting dan prehibridisasi pada metode ini dan latar belakang gel tidak menyulitkan pada pembacaan hasil.
