Abstract :

The aims of this study are to isolate and characterize E protein of Dengue-3 virus Surabaya isolates, therefore can be used as to produce of monoclonal antibody. This study is divided in some periods: isolation, characterization and analysis of E protein, the production of monoclonal antibody and to determine specificity of monoclonal antibody again E protein of Dengue virus in Indonesia from different strains. The result showed that molecular weight of E protein of Dengue-3 virus Surabaya isolate was 52 kDa, and it was antigenic protein that reacted specificity to a rabbit anti Dengue-3 polyclonal antibody. Fusion of B cell and myeloma cell produced 7 clones hybridoma that secreted monoclonal antibody again E protein Dengue-3 virus, which different specificity and affinity according to the type of immunoglobuline produced.
