Abstract :
The aim of this research was to determine and to manage the oocyte culture technique by conducting maturation and in vitro fertilization in local goat. Ovaries were collected at Pegirian sloughterhouse Surabaya and cumulus oocyte complexes (COCs) were aspirated from follicles in diameter of 2 to 5 mm. Oocytes were incubated in TC-199 medium with the addition of 5 IU and 10 IU Pregnant Mare Serum Gonadotropin (PMSG) hormone were compared to the control. All maturation were incubated at 38,5 oC with 5 % CO2 in humidified atmosphere for 24 h.The oocytes were exposed to sperm in EBSS medium with a final concentration of 12,5 X 106 cell/ml. Embryos were evaluated at 48 h after fertilization for cleavage. The percentage of embryo derived from maturation supplemented with PMSG was compared to the control by one-way anova analysis.In conclusion, oocytes maturation and embryos cleavage were significantly different (p
