Isolation, identification and purification of protein human menopause gonadotropin (hmg) from women menopause for follicle growth and in vitro maturation in dairy cattle

Abstract :
ABSTRACT The second phase of the research was aimed to determine the character of anti hMG. The characterization of anti – hMG was carried out through evaluation of specific reaction by using Indirect ELISA method. Booster injection was required order to increase titer anti – hMG, this injection was 100 µg or 2 ml protein of hMG in 0.2 ml Incomplete Freund Adjuvant (IFA). The optical density (OD) from ELISA with 1 : 10 dilution rate is described in the tables of attachement 3. As can bee seen from those tables that optical density (OD) of antibody against hCG increased from 1st week (0.576) to 3rd week (2.952) after first booster. In the 4th week the OD started to decline (1.971), and reaching the lowest point at 5th week (1.132). So, the second booster was performed in the fifth week, the OD then increased gradually in 6th week (2.148) and reaching the highest point at 7th week (3.177) and then started to decline again, the OD in the 8th, 9th and 10th were 3.070, 2.267 and 1.220 respectively. The production of anti hMG that was induced by hMG resulting from electroelution and measured by ELISA technique have shown that hMG from isolation of urine menopausal woman was capable to induce anti hMG. In conclusion, the result of characterization of hMG molecule protein from urine menopuse woman with molecular weight at 30 kDA has potentially imunogenic activity which capable of inducing humoral antibody against hMG (anti hMG)
